The process starts with a skin bioposy. The skin fibroblasts are then converted to Induced Pluripotent Stem Cells (IPSCs) using CRIPRa. Through this the dCas9 gRNA overexpresses the 4 Yamanaka factors (Oct3/4, Sox2, Klf4, c-Myc + LIN28A promoters) in the somatic cell reinducing their pluripotent stage.
The IPSCs are induced into primordial germ cell like cells (PGCLCs) by first turning the iPSCs into epiblast cells. The epiblast stem cells are pluripotent and will differentiate very early into primordial germ cells through the use of various culturing protocols.
Light microscopy is used as as a technique to proliferate the PGCLCs in vitro in an artificial tissue engineered environment. The scaffolds provide a favourable environment for cells adhesion, migration, and differentiation to facilitate the flow of culture medium to ensure the supply of nutrients and oxygenation to the cells. The PGCLCs will turn into immature eggs and sperm through their respective environments.
Dependent on the situation fertilization may occur through ICSI (Intracytoplasmic Sperm Injection) or IVF (In Vitro Fertilization). From there, although not required, Janam Labs highly encourages embryo testing (specifically PGT) in order to ensure that the best possible embryo is implanted into the uterus. Implantation would then occur through IUI, or simple embryo implantation.
